I also remember the criticism was that they couldn't be purified. Some people were very harsh with their criticism. I'm glad that time, science, and technology solved the issue.

Thanks for this great thread and your enthusiasm. I also started reading it today, and there are so many things I didn't know! Just one thing, it says transcriptional condensates contain TFIID and they cite the Lee et al. paper from Cissé lab, where this was, to my knowledge, not shown.

Ich wurde vom Netzwerk Chancen dieses Jahr nominiert und würde mich über eure Stimme freuen. Ich freue mich aber auch, wenn ihr für andere Nominierte abstimmt, denn es ist jetzt schon eine Ehre mit dabei zu sein. Es geht nicht um mich, sondern darum dem Thema größere Aufmerksamkeit zu verschaffen.

In conclusion, we propose that TAF2 partitions into different nuclear compartments, thereby linking transcription initiation and RNA splicing around nuclear speckles.

Does TAF2 have multiple roles in the regulation of gene expression? Yes. 1) TAF2 regulates promoters: Deletion of the TAF2 IDR leads to increased TAF2 promoter association. 2) TAF2 influences RNA splicing: Modulation of TAF2 protein levels and IDR deletion elicit alternative splicing events.

Why is TAF2 sequestered in nuclear speckles? We performed TAF2 proximity labeling and found that it interacts with splicing factors, such as members of U2 and U5-type snRNP complexes. The TAF2 IDR seems to modulate TAF2’s interaction frequencies with RNA-binding proteins.
