The dG thing is new and weird to me. But could a chemist analogy here be saying that breaking strained bonds (in cyclobutane or pick your favorite exemplar) "releases strain energy?" There is a problem in the formalism and the lure of "the line goes away" is too strong

You could do amine + CDI then benzyl alcohol (or alcohol first) if you are in a pinch. Shows up a fair amount for substituted benzyl variants

I find these papers somewhat interesting but mostly baffling (on top of the questionable description of synthesis). Are these numbers high? Compared to what? Should we aim to have a 100-mile pharmacopoeia?

I would kindly suggest a Sankey - designed to show flows - is not the appropriate way to represent this data.
