Our brains need a LOT of it! 🧠

We show that FLVCR2 transports choline into the brain & solved its structure in the OFS & IFS with choline bound. 🐀🔬

6/9 58 kDa MFSD2A was a little tricky to solve using cryo-EM by itself so we complexed it with a synthetic #antibody fragment from Tony Kossiakoff & @skerramilli at @UChicago

3/9 One wacky feature of this transporter is a disulfide bond that pins together the extracellular side of the N and C domains – could it be that this #MFS #transporter uses an adapted rocker-switch mechanism to selectively transport lysolipids?

2/9 We used #cryoEM #MolecularDynamics #FlowCytometry and #NativeMassSpectrometry to show that lysolipid substrates bind to a hydrophobic pocket in the MFSD2A central cavity, and are released into the inner leaflet of the membrane through a lateral gate in a Na+-coupled manner.
