We characterized the membranes after demineralization from pieces of shell from the edge of the opening.

Like other invert paleoproteomic studies, we were hindered by limited protein annotations. This study required conversion of several Conus genomes to protein databases with GeMoMa to have searchable gastropod databases.

As part of the study we also explored the mineral preservation with both scanning electron microscopy and Raman spectroscopy. We found that the membranes come from the calcite crust layer of the shells.

We detected the chitin with DART-MS not requiring addition sample preparation beyond isolating the membranes from the mineral matrix. To our knowledge, this is the oldest chitin detected by DART-MS.
