From old oyster mushroom, shiitake and lion's mane cultures in LB, 1mL was taken and put into fresh YPD media. The inoculated 20mL YPD in 50mL erlenmeyer flasks and 10mL YPD in 25mL erlenmeyer flasks were then aerobically incubated at RT.

Oyster mushrooms af 16 days incubation at RT showing near complete colonization on rye.
Next step is transfer to bulk media of wood chips and straw.
Soon testing YPD in 50 and 100 mL erlenmeyer flasks to grow aerobically. It should be more suited than the LB which was used before to reduce waste.

Promising growth of oyster mushrooms at 7 days incubation at RT on rye.
1:1 to 1:2 rye to water shows more/faster growth as compared to 1:2 to 1:4.

2 month old LB medium with oyster mushrooms was mixed slowly using a magnetic stirrer.
Using a P1000, 10 mL was put into fresh LB and 5 mL was added to rye substrate.
It is then incubated aerobically at RT

Rye was soaked for 24 hours in water and placed into boxes with air filters. Along with rye, 5:4 parts water was added after which it was autoclaved for 1 hour.

This will be used as substrate for different mushrooms.

Contaminated 48 hour growth of oyster mushrooms on YPD at 37C turned into a happy accident indicating possible antimicrobial properties.
Might do a later gram stain on the bacteria and attempt to further grow the oyster on a clean petridish.

Oyster mushrooms on YPD agar incubated for 24 hours at RT shows improved growth as compared to LB agar with the same conditions.

If you're growing microbes at home (or anywhere) it's most important to be safe. Follow the right protocol and research the waste flow beforehand. Always sterilize your plates before discarding them and you're tools for 15-60 minutes at high pressure.

Step 2: Colonies were cut with a sterile scalpel and put into YPD (1% yeast extract; 2% peptone; 2% glucose w/v%) and incubated at RT.

Note: YPD is more suited for fungi, a cheaper alternative would be PDA, for instance from doi 10.1111/j.1574-6968.2007.00923.x or the Merck Millipore protocol

First steps:
1.5% LB agar and liquid LB media (1% peptone; 0,5% yeast extract; 1% NaCl w/v%) inoculated with shiitake, oysters and lions mane on 28-02-2025 from mycelium syringe in sterile environment using sterile water as control on plate
Incubated at around RT
NOTE: YPD is probably better!
