Our structural and mechanistic analyses reveal that the TM domain of IST2 scrambles lipids in the ER membrane, whereas its flexible tether provides a scaffold for the lipid transfer protein OSH6 — another example of how scramblases may support non-vesicular lipid transport at membrane contact sites.
