The microtubule links between spindle poles and the apicobasal cortex were strongly reduced after F-actin disruption (LatB), correlating with spindle tilting. This suggests F-actin helps anchor the spindle to the cortex once the PPB has disassembled.
August 25, 2025 at 9:21 AM
In WT plants, expansion microscopy with spindle immunolabeling showed microtubule connections between spindle poles and the apicobasal cortex.
August 25, 2025 at 9:15 AM
F-Actin maintains spindle orientation against cellular geometry.

In WT, spindle poles aligned with the PPB, while LatB-treated and act7-4 cells showed spindle misalignment at metaphase.
August 25, 2025 at 9:06 AM
Actin perturbation disrupts the alignment between the PPB and the division site.

Time-lapse analysis of dividing trichoblast in root expressing the microtubule reporter IQD8-GFP (green) and the plasma membrane marker 2xmCh2xPHFAPP1 (magenta), after 2h-LatB.
August 25, 2025 at 9:03 AM
Actin perturbations reset division orientation toward geometry-driven mechanisms:

Actin provides essential spatial cues that reinforce robust patterning, particularly when geometric constraints are not aligned with growth direction.
August 25, 2025 at 9:00 AM
In WT roots, divisions were mainly transverse anticlinal, whereas actin disruption caused abnormal longitudinal or periclinal divisions.
August 25, 2025 at 8:57 AM
PPB-defective mutants (ton1a, trm678) show tilted but laterally confined divisions, suggesting safeguards prevent apicobasal division

F-actin (cyan) accumulates at apicobasal cell faces during division, supporting a potential instructive role for F-actin in orienting the division plane
August 25, 2025 at 8:55 AM
The preprophase band (PPB) sets division orientation before mitosis.

In the absence of PPB, in the trm678 mutant, division planes in atrichoblasts no longer followed the shortest geometric path but appeared randomly anchored along the lateral surface.
August 25, 2025 at 8:50 AM
Errera’s rule states that plant cells tend to divide along the shortest path, splitting the cell into two equal volumes, much like a soap bubble dividing into two smaller bubbles:

Here, we found that root epidermal cells do not systematically divide according to area minimisation
