13/ By tracking individual cells we saw that histone levels do not change prior to release from HU and are at mitotic levels. Thus, histone synthesis completed while DNA remained unreplicated.

12/ By flow cytometry we could see DNA replication proceed following release from HU, but histone levels were constant.

9/ At this point I wanted to look at histone dynamics in H. uvarum itself, so we inserted a tagged H2A marker to track histone levels during the cell cycle using time-lapse imaging. Thanks to Jürgen Heinisch for kindly providing all transformation reagents and protocols.
