Based on our results we propose that macrophages modulate the intrapituitary hormonal balance, at least in part, through extracellular ATP-mediated intercellular signalling.

While Microglia-like pitMØs localize in the posterior pituitary, other pitMØs home to the anterior pituitary and interact closely with hormone-secreting cells.
By carefully depleting these macrophages, we observed a reduction in specific intrapituitary hormone concentrations.

Using scRNA-seq, fate mapping, and imaging, we show that the pituitary gland hosts several specialized types of macrophages (pitMØs) with distinct developmental kinetics and characteristics.
All pitMØs originate from yolk sac progenitors and self-renew independently of BM-derived monocytes.
