We found 4 proteins-cancer associations that had evidence from all analysis performed
January 21, 2025 at 1:59 PM
Out of the 618 - we wanted to identify if there was any genetic evidence to support these associations -> We used cis-pQTL but also created exome scores (projected into the full ukbb exome data). We found strong evidence for one cis hit and 28 proteins using the exome-scores

We identified 182 proteins that differed <3 years before diagnosis --> providing a potential marker for immediate cancer
January 21, 2025 at 1:43 PM
when we stratified out into < 3 yrs to diagnosis, 3-7 yrs and > 7 yrs (median time do diagnosis in this group was 12 yrs). In the >7 yrs group we identified 107 protein-cancer associations.....
January 21, 2025 at 1:37 PM
Using the invaluable @uk_biobank and the @OlinkProteomics data, we investigated 1,463 proteins across 19 common cancers, finding 618 protein-cancer associations.... (Thanks Trishna Desai for this great figure 1)

My sister (aged 41) is terminal with metastatic breast cancer. I was told the Peter Mac was the best in Australia. Tried to get a second opinion to explore all treatment options here in Oz. No phone call. This is how the Peter Mac treats the most desperate cancer patients.

We just had a very exciting unit seminar talk by @ellvlirae from the Genomic Epidemolgy Branch(GEM) at @IARCWHO on the largest GWAS results from the @HEADSpAcE_Study

Today is my last day @IARCWHO. My top highlight was having my concept selected for the cover of Nature Genetics cheers mutographs team. Looking forward to my next big adventure at the @UniofOxford in the new year. But now, enjoying some downtime with my family back in Australia

This shows good evidence that smoking behaviours are influenced by the individual's germline genetics which influences their lifetime carcinogenic exposure -> increased somatic mutation burden

As expected for the CHRNA3-5 hit that we call Mr Big, the association was stronger and additional independent associations were discovered

Another novel finding is in 15q24 near CYP1A1 and CYP1A2 - This finding colocalises with lung function but not with SMOKING TRAITS - may be related to air pollution ????? - more work is need to explore this cool finding...

By meta-analysing these results, we have discovered an additional 8 new loci (coloured in red).......
January 21, 2025 at 1:43 PM
We know that family history of lung cancer is a susceptibility factor - so we performed a genome-wide association by proxy (GWAx) within the @uk_biobank and compared this to the traditional GWAS approach using LC cases, the Manhattan plots are very similar ...
