Nah you can balance one tube, just need to tape it to another centrifuge spinning the opposite direction

You could try sequencing TN7L and TN7R sites to make sure nothing is wrong there. It can also be very inefficient with larger genes - sometimes its good to make 4-5 plates for each construct for these.
