A little bonus take-away: we found that #BUSCO is inaccurate in these conifer megagenomes, many of the predicted single-copy genes actually show considerable variation in normalised coverage. It is understandable as up to odb_10, no reference species is a conifer.

Our study demonstrated how long-insert #sequence capture and a de novo local assembly approach help study genomics effectively in these #megagenomes (> 20 Gbp)! Also a big shout out to @ArborBio and @nanopore!

Interestingly the gene involved in the pathway one step before, Ugt5b, is found to be dominantly single-copy.

This gene encodes the enzyme that catalyses the conversion of glucosides to the active forms piceol and pungenol, which confer #resistance against herbivory of spruce #budworm.
