A Marshall just shouted “only 3 hours left!” And the look I gave him did this.

Lap 4 update: oh no

Currently done lap 3 and has gone exactly as above, with a major crash due to overexcitement. (Bike and rider are fine)
I plan to learn nothing from this.

My usual performance goes:
Lap 1: take it slow, pace yourself.
Lap 2: why is everyone so SLOW? I’m bored, imma overtake this group, and the next…
Lap 3: imma do a hot lap!
Lap4: oh no. I’m exhausted. How long has that been? [hour 2 of 7]. Oh no.
Lap 5: screaming, crying, throwing up

Thats where the story ends at the moment - we would love to look at the in vivo effects of HpBoRB against RELM-beta (does it suppress RELM-beta's activity?) but we ran out of time in Viv's PhD. If anyone has any interesting RELM-beta assays they would like to try HpBoRB in, hit us up!

We then confirmed this binding in a number of ELISA, SPR and gel filtration assays - it is a high-affinity (subnanomolar) interaction.

We screened the CCP domain protein family to see if there were other host immune factor binders, using an AVEXIS protein-protein interaction screen. The screen confirmed that HpBARI binds ST2, and that TGM1 binds TGFBR2 (reassuring) and identified one new hit - a protein which bound RELM-beta.

This story started when we realised that all of the immunomodulators we had identified from Hpb (HpARI, HpBARI and TGM) were related in their domain structure - all consist of strings of CCP domains. Turns out the CCP domain family is massively (and uniquely) expanded in the Hpb genome.
