Looking at associations at non-linear order, for these genes whose expression is always correlated with mechanics, we could observe both sigmoid and band-pass filter (!) behaviour for gene expression in response to changing mechanical forces:

But spatial mechano-transcriptomics also allowed us to ask a more general question: what is the relationship between the gene expression profile of a cell in a tissue and its mechanical state? This led to the discovery of 'mechano-associated' genes:

Using spatial gene expression profiles, we were able to unravel the molecular basis of this biophysical phenotype. Ephrin ligand-receptor interactions control mechanical tension via cell-cell adhesion and cell contractility for cells at tissue compartment boundaries!

We observed that individual tissue compartments do not have a unique mechanical profile, but that an increased mechanical tension is always observed at the boundary between two different cell types:

Using this approach, we were able to study the formation and maintenance of tissue compartment boundaries in the E8.5 mouse embryo:

Using image-based spatial transcriptomics (seqFISH, CosMX, Xenium, etc.) and cell shape-based mechanical force inference, spatial mechano-transcriptomics allows simultaneous spatial profiling of gene expression and mechanical forces at single cell resolution.
