Overall, our study provides a reference for future research on #aged oocytes and from #infertility #patients. With our previous work, our results further highlight how oocytes from different mammalian species adopt different strategies to regulate #proteostasis during their extended #life.

And what about protein #aggregates? While mouse oocytes store them in large endolysosomal vesicular asseblies (ELVAs), human oocytes seem to sequester them inside individual enlarged lysosomes (similar to what some quiescent stem cells do).

In most MIIs, mitochondria also start to surround clusters of ER membranes below the cell cortex. Another super-organelle awaiting to be studied maybe?? 🤩

We also found that organelles undergo a dramatic redistribution from GVs to MIIs: while #lysosomes undergo exocytosis, as highlighted by the appearance of LAMP1 on the cell surface, #mitochondria spread throughout the cytoplasm

We then compared GVs and MIIs from the same donors. Unlike in #mouse oocytes, the labelling intensity further decreased in human oocytes from GVs to MIIs, suggesting a general downregulation of organelle activity during oocyte maturation.

We first compared oocytes with the surrounding somatic cells (cumulus cells, CCs). We found that the intensity of all dyes is lower in oocytes compared to the adjacent CCs, suggesting lower organelle activity in the germline.

We systematically surveyed changes in organelles' activity and localisation during human oocyte maturation by labelling both immature oocytes (GV) and mature eggs (MIIs) with fluorescent dyes for lysosomal, proteasomal, and mitochondrial activity, and imaging them live.
