In Animals, TFs have a clear preference for certain chromatin states. With some tricks, we show that this could also be true in plants. This opens the possibility to distinguish TFs by their mode of action (pioneer vs normal TFs). This includes some unusual 5'UTR TFs.

Chromatin is highly conserved across eukaryotes, but it does not always work the same way. First, in both Arabidopsis and Marchantia, analogous chromatin states can be identified. Tissue-specific genes associate with TFs in facultative chromatin.h

Marchantia has probably the most simple cell cycle control system ever described in plants and beyond. This has several evolutionary implications, it could help to unravel the system in flowering plants and find out how this system may work in ancestral eukaryotes.

Using overexpression, we characterized how these cyclins affect cell division. We found that MpCYCD;1 is the rate limiting cyclin; MpCYCD;2 is not functional; and surprisingly, MpCYCB;1 and MpCYCA arrest cell division. This means that they need to be degraded to complete the process.

Is this true in vivo? We did translational reporters and validated this observation in vivo. Interestingly, CYCB is accumulated in the cytosol, but for a few minutes, it is translocated to the nuclei to start mitosis.

During cell-cycle re-entry, only one of each cyclins is activated at a time. Looking at single cell data during progression, we manage to separate subclusters of three clear phases corresponding to G1, S, and G2/M. We find out that each cluster is governed by CYCD1/CDKA, CYCA/CDKA, and CYCB1/CDKB.

For example, in Arabidopsis you have 30 cyclins all over the place… Marchantia has a small set of cyclins, including 2 CYCD, 2 CYCB, and 1 CYCA. Phylogenetic analysis suggest that these genes are single copy in most related non-seed plant species too. (Why so many cyclins in flowering plants?)

In Eukaryotes, to coordinate replication with cell division, an elegant relay of cyclins and CDKs controls the transition between gap 1 (G1), replication (S), gap 2 (G2), to end in mitosis and start over again. You may have seen this plot in the Alberts manual, but is it true in any organism?
