In this concept:
• A signal peptide directs the translated transporter into microsomal lumen in an inside-out orientation
• Cytosolic loops are stabilized by co-expressed synthetic nanobodies (screening also by CFPS)
• Auxin substrate is added to enhance folding efficiency during translation
