We also observed OBP bound to a DNA hairpin with a single-stranded tail, an intermediate state in the DNA unwinding. Here OBP switches from dimer to monomer, revealing dramatic conformational flexibility during origin activation.

Furthermore, we can resolve a long-standing paradox: why C-terminal deletions enhance helicase activity but reduce replication efficiency? It turns out the C-terminal tail threads into the partner monomer in proximity to the ATP-binding pocket, regulating ATP hydrolysis and helicase activity.

We can show how the conserved RVKNL motif binds sequence-specifically to the HSV-1 origin of replication OriS. Our structures reveal that Lys758 and Arg783 are responsible for the discrimination between the three recognition sites (Box 1, 2 and 3) within OriS.

Using cryo-EM, we determined OBP structures at up to 2.8 Å resolution in multiple states:
1. bound to HSV-1 origin of replication (OriS) DNA

2. with a non-hydrolyzable ATP analog

3. as monomer, dimer, and higher-order assemblies
