Finally – does this matter to patients? Yes! CINMN high, myeloid-infiltrated tumors have poor clinical outcomes – meaning we need new treatments for these patients. 9/11

We also identified tumor cell-intrinsic CXCL8 expression in CINhigh EAC, potentially accounting for the myeloid-dominated tumor microenvironment (Melero lab, de Andrea lab @unav.edu) 8/11

Using snRNA-sequencing (Izar lab @columbiauniversity.bsky.social) with matched multiplex immunophenotypic profiling (@jleslie1.bsky.social) we found tumor cell-intrinsic innate immune activation and intratumoral myeloid infiltration as phenotypic consequences of ongoing micronucleation in EAC. 7/11

Using tumor samples of EAC, we developed and validated scoring of cGAS-positive micronuclei as a measure of ongoing CIN. Excitingly, this simple score neatly matched orthogonal measures of CIN in WGS (thanks @kcakdemir.bsky.social & team!) 6/11
