We’re welcoming Emma Andretta back to the magnificent world of #gdTcells! Before joining the Master’s program at Mount Sinai, she learned about the #bestimmunecellever with @boysonlab.bsky.social and then expanded to mucosal immunology at MSKCC in the Rudensky lab. Welcome Emma! 🤩

She’s got her white coat, so now @ananya-p.bsky.social is an official PhD student @sinaiimmuno.bsky.social ! That’s how that works, right? More #gdTcell magic to come! 😁

Despite having more γδ IELs, γδHYP mice do not exhibit overt pathology. We find that this is because the mature CD39hi γδ IELs are more tightly regulated; they proliferate less, and are less likely to degranulate or produce less pro-inflammatory cytokine in response to stimulation. 9/n

It's been known for a long time that IL-15 promotes γδ IEL survival, and here is no exception as cell survival is increased in γδHYP IELs. As @mahimaswamy.bsky.social showed, IL-15 also promotes γδ IEL protein production & we find increased mitochondrial metabolism in CD39hi γδHYP IELs. 8/n

Further upstream, treatment with broad-spectrum antibiotics is sufficient to reduce CD122 and CD39 in γδHYP IELs, but it doesn't affect enhanced epithelial IL-15 production or the number of γδ IELs. This suggests that sustained γδHYP microbiota exposure is not req'd for CD39 upregulation. 7/n

TCR activation in the naive CD39lo γδ IELs is also critical for driving both CD122, and subsequently, CD39 expression as there is an additive effect on its expression when you culture CD39neg γδ IELs with both stimuli. TCRγδ internalization prevents the upregulation of both CD122 & CD39 in vivo. 6/n

CD39hi γδ IELs expressed the highest amount of CD122 and if we cultured CD39neg γδ IELs with IL-15RC, we could convert them to CD39pos or further upregulate its expression in already positive cells. Anti-CD122 blocked the upregulation of CD39 in γδHYP mice. 5/n

To determine why CD39 expression was increased, we performed CITE-seq and found that CD39 expression positively correlated with transition from a naive-like to tissue-adapted, mature γδ IEL state. Moreover, CD39hi γδ IELs expressed genes downstream of TCR and IL-15 signaling. 4/n

Bulk transcriptomics revealed an increase in Entpd1, the gene encoding CD39, a known marker of regulatory γδ T cells. We found a clear shift in CD39 expression in γδHYP mice with more CD39hi γδ T cells in phenotypic mice. High CD39 expression was a feature of natural IELs, but we focused on γδs. 3/n

I debated posting this publicly, but wanted to acknowledge Dr. Alyssa Lillian’s passing and what she meant to us. Alyssa (A2) was part of our lab family and her smile, snark and curiosity were a joy to us all. Our thoughts are with her family, friends, Rutgers NJMS & Hopkins colleagues 💜

Great night out at #NYC @crohnscolitisfdn.bsky.social #TakeStepsWalk with the lab! The Mount Sinai IBD Center raised almost $2,500 for #IBD research. Bonus: I met Blake Lively & Ryan Reynolds (both very nice) on the ferry ride home! @sinaiimmuno.bsky.social

An added bonus of #DDW2025 was hanging out with @aaronblocker.com and Megan Starshak, #IBDsky patient reporters that I’ve known for years through our community 🤩 Patients need to hear why #GIresearch is so important so that they can @standupforscience.bsky.social!
