It's possible that there may be a way to modify the exp3.json files directly and have VTube Studio reload them, but the modification therein and reloading the files isn't really within my realm of knowledge, and I don't know if it would work. Sorry I can't be more helpful!

Unfortunately, on reviewing the documentation for VTube Studio's API, it doesn't appear that there's an endpoint for anything to do with Expressions other than getting Current Expressions/States or requesting the full activation/deactivation.

Right, I gotcha. That makes sense. Initial thoughts are that this should be doable, possibly without the use of custom C# if we can get the Send Raw Request to work with this. When I have some time, I'll try and do some testing, but here's the doc page for now!

Is this in reference to like, VTubeStudio toggles? That specifically isn't something I'm super familiar with. It's possible that Streamer.bot may not have the functionality you're looking for out of the box, but it might be attainable using the VTubeStudio REST API via custom C#.
