5 layers is pretty unlikely. Manufacturers prefer symmetric PCBs with an even layer number to use standards stackups and manufacturing processes.
Cores are pre-assembled with two copper foils and manufacturers just stack them with prepregs

That's an other point. Why would a designer request for glue here ?
Some are far two conservative or think of glue for wave soldering while the product is reflown (and they forget other wave soldering needed features)
That's a question to ask to the designer team

You have to specify where you want glue, but also where you don't want any.
