Because the green and red monomers both dimerize with the B monomer, Contact-FP can also be used to visualize 2 contact sites that share an organelle, e.g., ER-LD and LD-mitochondria 9/11

Contact-FP can be used to visualize dynamic interactions between any organelle pair represented in the toolkit – shown here for LD-mitochondria. 8/11

We engineered a suite of ddFPS (red, green, and B monomers) targeted to 8 compartments: cytoplasm #LipidDroplet #ER #Mitochondria #Peroxisomes #PlasmaMembrane #Caveolae and #Lysosome. 5/11

We turned to a palette of dimerization-dependent fluorescent proteins (ddFPS) developed by Robert Campbell’s group. ddFPs consist of two weakly fluorescent monomeric proteins that reversibly interact to form a fluorescent heterodimeric complex. 4/11

Membrane contact sites allow cells to channel ions and lipids between compartments. We know from previous work that membrane contact sites are very dynamic. These images show lipid droplets transiently interacting with up to 5 other organelle types over a 5-minute period. 2/11
