Our findings reveal a compartmentalized system in which presentation of dietary vs pathogen-derived antigens is accomplished by distinct APC subsets, likely as a mechanism to prevent food-specific Th2 responses even amid strong type-2 immunity driven by helminth infection.

Surprisingly, these inflammatory cDC2s did not engage in dietary antigen presentation and thus failed to induce differentiation of diet-specific Th2 cells, avoiding food allergy development.

We found that S.v. infection impairs oral tolerance induction by excluding tolerogenic APCs from antigen presentation to food-specific CD4+ T cells, while inducing a large population of inflammatory cDC2s, which promote Th2 responses.

We previously discovered that infection with the helminth Strongyloides venezuelensis (S.v.) prevents Treg and oral tolerance induction. With this information in hand, we asked what happens to the APCs presenting dietary antigens during concomitant helminth infections.

To tackle this, we adapted LIPSTIC technology to directly identify, characterize, and locate APCs presenting food antigen in vivo. This allowed us to pinpoint which cells regulate food-specific tolerance vs inflammation.

There are many different types of APCs and if you think about the high density and diversity of luminal antigens constantly present in the gut, identifying which APC directs a specific immune response in the intestine is highly challenging.
