-Restart your game for good measure. There won't be any way to confirm if it's working other than starting the game, selecting the custom character you desire, and seeing if the second consecutive power up you pick up is a mushroom. If it is, congrats! It's working. If not, something needs fixing.

-Put both characters that came with the download into the "custom-characters" folder. You can find this by launching SMBR1, going to custom levels or resources and select the "open folder" option. Go back one folder from the option you chose, and there should be a folder labeled "custom-characters."

I got it to work by doing the following:
-I made a "mods" folder in the folder with the "SMBR1" application, and put the unzipped "someoneuknow9097-SMB1R_WG" file into that folder. Don't confuse it with the "mod-hooks" folder that will be created when you make the "mods" folder.

That's good to hear! Making comics can be a huge investment, and motivation can be an issue with larger projects, so it's nice to hear that you're still having a good time.
