Finally, fantastic Vincenzo Mastrolia used mice expressing Channelrhodopsin in Chandelier Cells, the interneuron forming axo-axonic synapses, to demonstrate that this nanoscale reorganization was associated to a strong reduction in evoked IPSC

However, when looking at Gephyrin sub-synaptic domains, the story is different.
Interestingly, only Gephyrin SSD at the AIS were reduced in volume, while those at tufts remained unchanged. Also, not only AIS SSDs on hM3Dq+ neurons, but also neighbors expressed this plasticity.

cfos activity increased as the network level and AIS length shrinked as expected from previous work. However, in contrast to when increase activity happen at P12-P18, no change of axo-axonic synapse number occured here.

Literrature is solide about the link between Gephyrin and GABAa Receptors clustering. Still, we checked in cell culture that both Gephyrin and GABAaR SSD properties correlates. We used Abbelight spectral demixing feature to perform dual color SMLM. (Thank you @nibourg-abbelight.bsky.social)

Here we took advantage of Gephyrin-FingR from @Don_B_Arnold to label in vivo endogenous Gephyrin. We noticed that depending of the position of inhibitory synapses, hence the type of interneuron forming synapses, Gephyrin Sub-Synaptic Domains had specific properties

We first challenged the possibility to image inhibitory synapses at 2 compartments of S1 L2/3 Pyramidal Neurons using Single Molecule Localization Microscopy in brain sections. ✅

We Increased network activity in vivo and used dSTORM to look at the nanoscale organization of inhibitory synapses formed onto the AIS and onto tufts dendrites of L2/3 S1 Cortex!
