I believe my lab did test before my time here different backbones of which a phosphorothiote ones were included. Will ask my PI what he thinks

We have shown that modification of an siRNA strand at position 1 and 2 by 2’-OMe, we can prevent its loading.
Yes our lab is currently working on developing the concept of 6mer seed toxicity as a therapeutic.
