We extracted neuronal enriched proteomes across the length of DRG sensory neurons, giving insight to the compartmental specificity from peripheral to central terminals.

While neuronal signatures are detected in each compartment, they also show functional specificity, as expected. Primary afferents synapse on secondary order neurons in the dorsal horn of the spinal cord. This corresponds to the enrichment of proteins involved in presynaptic function and synaptic signalling between neurons, as well as vesicle-mediated transport, exocytosis, and other synapse-related terms in the LSC. The DRG contain neuronal somata, and, thus, corresponding functional enrichment matches expectations. Here, we see the enrichment of proteins involved in mRNA processing, post-translational modification, and nuclear structures, in line with the biosynthetic activity expected in neuronal cell bodies. The sciatic nerve consists of neuronal axons, as well as Schwann cells, fibroblasts, endothelial cells, and immune cells. Its neuronal proteome is enriched for myelination, cytoskeletal organization, and vesicle-mediated transport. In the “myelination” terms, we see the presence of nodal and paranodal proteins such as Cntn1 (Contactin-1) and Cntnap1 (Caspr1), as well as more generally expressed neuronal proteins like Ncam1, Nefl, Nefm, and Nefh. We also see proteins more classically associated with myelin itself, such as Plp1, MBP, and MPZ , which also show expression in neuronal subtypes.
