In this article, the authors present a spatial multi-omics atlas of TLS-positive pancreatic ductal adenocarcinomas from patients treated with neoadjuvant immunotherapy, revealing that pathologic responses are associated with intratumoral TLS that support local maturation of IgG-secreting B cells.

Inadequate CAR T-cell infiltration into solid tumors limits efficacy. The authors show that engineering CAR T cells to express osteopontin can overcome physical barriers in solid tumors, offering a promising strategy to improve CAR T-cell infiltration and efficacy.
