Finally, we compared mice to humans and saw conservation of most neuronal types, but divergences in neuronal proportions and transcriptomes, in particular for cell surface receptors. If you want to do drug discovery, these details matter!!

We then went to town on the glial populations where we identified some pretty profound sex differences. We did not note sex differences in neurons, but the identification of male and female specific glial populations for astrocytes, microglia, and oligodendrocytes could have important functions.

And, of course, we did the same with the ventral horn, giving confidence in our identification of motor neuron pools.

We used this information to create detailed maps of the locations of cell types in the dorsal horn. This gives great insight into sensory/nociceptive circuits in the human dorsal horn with a resolution that has not been seen before at this scale.

Of course, we also care about non-neuronal cells and the spatial arrangement of all of these cell types so we used Xenium spatial transcriptomics to map the locations of these cell types using gene markers from our single nucleus work. Our custom panel led to many important insights.

We did the same thing for ventral horn neurons (we microdissected the dorsal horn from the ventral horn to have clear sequencing) and were able to clearly delineate motor neuron populations.

We mapped dorsal horn neurons using single nucleus sequencing and developed detailed transcriptomes for subtypes which we named using a nomenclature system based on work done at the Allen Brain Atlas.

We did single nucleus sequencing and Xenium @10xgenomics.bsky.social spatial transcriptomics on male and female spinal cords from organ donor recovered tissues in collaboration with Southwest Transplant Alliance. We started this work about 5 years ago. We optimized and optimized and optimized.
