Its* . Heads up make sure your DAPI is set up to catch the emission from 421 - a lot of DAPI filtersets cut on too long and chop off a lot of the signal.

Don't sleep on flow dyes - BV421 (the donor at the base of most of the Brilliant Violets) and it's analogues are very bright and photostable. A little more inclined to clumping than most, but surely you're already spinning your tubes and pulling off the top.

Why waste a perfectly good protein/antibody channel on some crappy nuclei. I KNOW I'm looking at cells!
