Behaviourally we observed:

1) females showed more characteristics of distress with fewer long calls and more calls overall
2) females showed difference in total distance moved and time spent in the center of the open field

Mouse brain development showed:

1) Larger ventricles during the gestational period.
2)Show decelerated brain growth in early life
3) Females show lower brain volumes and growth in key regions like thalamus and amygdala

Mice were

1) smaller during gestation
2) underwent accelerated growth (as indexed by weight) after they are born
2) female mice stay underweight once they reach adulthood and male mice catch up

We examine three cohorts of mice:

1) At late fetal stages
2) Early on in post-natal development
3) After weaning all the until post-natal day 90

We used a combination of maternal observations, stage appropriate behaviours, and longitudinal MRI postnatally

As a positive control we injected the hippocampus and found that what held true for the striatum doesn't hold true here. We don't see wide-spread neurodeneration and the SIR model does not predicit outcomes. Overall, this suggests differential vulnerability throughout the brain to a-syn propogation

In collab with @alaindagher.bsky.social @misicbata.bsky.social and Shady Rahayel, we used the SIR model to computationally model neurodegenration as a function of the structural connectome and gene expression. SIR cpatures the dynamics better than axonal connectivity (very bad!) and gene expression!

Using MRI we tracked the neurodegeneration across these mice over time and see a wide-spread neurodegnerative profile over time that matches previous observations from our studies. The neurodegeneration also propogates across the connectome of the brain. M83 receiving mice fibrils are worse

Regardless of the PFF-type or genotypes we see motor impairment, weightloss and poorer survival. This is worst for the M83 models that receiving mice PFF (as expected) but suggests there's a general mode of spreading across genotypes and species.

Lifestyle is differentially related to the relative contributions of different microstructural measures in a sex-specific way. We also find composite signatures related cognition is as well. This suggest that our methods capture brain variation that may go undetected using trad methods.

We used OPNMF to derive spatial groupings of hippocampal microstructure (FA, MD, T1/T2) that replicate, to a great extent, the findings in our 2020 paper, suggesting this organization is a stable human phenotype in the human hippocampus:
