CRISPRs on plasmids are great in defence, but in offence limited by TA – this helps explain how bacterial immune systems act & informs use of CRISPR technology.

Great, but what about the real world? @leightonpayne.bsky.social bioinformatically analysed plasmid-borne CRISPR-Cas systems and the plasmids they target. When we look at (nucleolytic, non-Type IV) plasmid-borne CRISPR-Cas we find that they try to avoid targeting plasmids with TA!

And this is also what we see experimentally! Defensive or co-invading CRISPR-Cas are always beneficial, but offensive CRISPR is severely limited by and even detrimental when there is TA.

First, we conceptualise: modelling by @brownlab.bsky.social predicts that resident CRISPR-Cas usually benefits its plasmid (higher competitive outcome), but that CRISPR can actually be a liability when it is offensive & the competitor plasmid has TA! (lower competitive outcome)

Immune systems on MGEs are so cool!
We know that plasmid-borne CRISPR-Cas are active in plasmid competition, we know that toxin-antitoxin systems are prevalent on plasmids, and we know that they interfere with each other – so why have CRISPR on a plasmid?
