Adipocyte-derived extracellular vesicles are key regulators of central leptin sensitivity and energy homeostasis
The exact mechanisms underlying leptin resistance remain elusive. Here, Wang et al. report that adipocyte-derived EVs and their miRNA cargo are key regulators of central leptin sensitivity and that the loss of these leptin-sensitizing miRNAs in adipocyte EVs contributes to leptin resistance and obesity.

Induction of a metabolic switch from glucose to ketone metabolism programs ketogenic diet-induced therapeutic vulnerability in lung cancer
Zhengwei et al. show that lung tumor-initiating cells (TICs) adapt to glucose limitation by switching to ketone utilization, dependent on MCT1 and its chaperone CD147. The ketogenic diet expands the TIC pool yet, paradoxically, induces vulnerability toward MCT1 inhibition, demonstrating how dietary intervention may be exploited to enhance anti-cancer therapies.

Quercetin-derived microbial metabolite DOPAC potentiates CD8+ T cell anti-tumor immunity via NRF2-mediated mitophagy
DOPAC, a microbial metabolite of quercetin, enhances CD8+ T cell-mediated anti-tumor immunity by activating NRF2-driven mitophagy, thereby improving T cell fitness and potentiating the response to immune checkpoint blockade.

Dietary urbanization destabilizes host-gut microbiome homeostasis and informs precision nutrition for human health
Dietary urbanization has reshaped the human gut microbiota relating to the rising non-communicable diseases. Zhang et al. deconvolute the impacts of urban diets on the gut microbiome leading to non-communicable diseases and propose a microbiome-directed precision-nutrition framework to combat diseases.

The enedioic acid analog 326E alleviates metabolic dysfunction-associated steatohepatitis via dual targeting at ACLY and PPARα
Xie et al. demonstrate that 326E, an enedioic acid analog, safely and effectively reverses MASH in multiple preclinical models by dually inhibiting ACLY and activating PPARα, with early clinical trials showing good tolerance and potential for further development.

Machine-learning-guided discovery of SLC25A45 as a mediator of mitochondrial methylated amino acid import and carnitine synthesis
Khan et al. apply machine learning to identify substrates of UNC93A, SLC45A4, and SLC25A45. Biochemical assays and in vivo metabolite tracing establish SLC25A45 as a key mediator of mitochondrial import of methylated amino acids and carnitine synthesis. Loss of Slc25a45 impairs fasting response by limiting carnitine availability for β-oxidation.

Paternal exercise confers endurance capacity to offspring through sperm microRNAs
Yin et al. show that paternal exercise improves offspring endurance capacity and metabolic health via sperm microRNAs that reprogram gene expression in early embryos, revealing how exercise benefits can be inherited across generations.

Catenibacterium mitsuokai promotes hepatocellular carcinogenesis by binding to hepatocytes and generating quinolinic acid
Zhang et al. reveal that the gut bacterium Catenibacterium mitsuokai promotes hepatocellular carcinoma by disrupting the gut barrier, colonizing HCC cells via its surface protein Gtr1/RagA and receptor γ-catenin, and secreting quinolinic acid, which binds to TIE2 and activates the oncogenic PI3K/AKT pathway in HCC cells.
