And finally, we generated an interesting mutant in Cep83: the mutant lead to the exclusion of one exon that lead to loss of basal rate of ciliogenesis in mouse ESCs. But in gastruloids, ciliogenesis was restored.

But centrosomes are needed during gastruloid differentiation. But this can be rescued by additional deletion of p53.

Only when the cells were placed into the right 3D environment, primary cilia were induced. Furthermore, cells without primary cilia can differentiate well, albeit with a slight reduction in symmetry breaking during gastruloid differentiation.

Isa generated numerous KOs of either centrosome or cilia components and analyzed their role in the exit from naive pluripotency, in in vitro rossette assays and in gastruloids. On the way she discovered that simply differentiating the cells does not induce ciliogenesis.
