🧐In a nutshell: CK1-mediated multisite phosphorylation of Dishevelled dramatically alters its bulk electrostatics, promoting DVL intramolecular coupling between the positively charged cleft of the DEP domain and the adjacent phosphorylated disordered region.

📌AH078 is a powerful tool for studying CK1 δ/ε biology in cells, enabling rapid depletion of both kinases—something challenging with KO systems due to cellular dependency on CK1 δ/ε activity!

👉 The PROTAC AH078 is characterized through in vitro and cellular assays, demonstrating on-target activity, selectivity, and a VHL-dependent mechanism of the CK1 degradation in cells.
