... 2️⃣ Logic-gating: IF-BETTER gates target 2 antigens-> enhanced activity when primary Ag expression low 3️⃣ inactivating mutation of 2/3 of CD3 ITAM domains improves CAR-T cell persistence w/o affecting function - only 25M dose sufficient for protective response in B cell lymphoma

Also on #IUIS2025 Day 3: keynote by Ruslan Medzhitov. Pet shop mice have pre-existing immunity (look at the size of those LNs!) and a crowded "antigenic space" with Th1/17/reg immunity. Allergic sensitization in SPF mice is possible because the antigenic space is available for full blown Th2

Later on: @victora.bsky.social at RockefellerU used brainbow mice to study BCR evolution in individual GCs - tl;dr: GCs mostly do mutations that are easy, not most enhancing/ GCs are intolerant to loss of affinity / clonal bursts are not main drivers: rather slow evolution biased to higher affinity
