6️⃣And not just in🧫!! We confirmed BMP upregulation in P14 🐁!! Strikingly, when we used a specific BMP inhibitor around P14 🐁, we partially rescued the over-activated neurogenesis seen in P30 Sox5 mutants. This stresses the role of BMP signalling in the acquisition of the right quiescence deepness!

5️⃣To confirm this, we studied NSCs in vitro 🧫. Here, we found that Sox5 prevents excessive activation of the BMP canonical signalling pathway, which we have now determined is associated with the shallow quiescent state in this model.

4️⃣To dive deeper🤿in the changes that occur during the critical window of P14 at the #Hippocampus, we performed transcriptomic analysis of Sox5 mutant NSCs. This analysis revealed dramatic changes in developmental programs, including an upregulation ⬆️ of the BMP signalling.

3️⃣When Sox5 is lost, there is an accumulation of shallow quiescent NSCs at P14 more prone to activation. This causes a neurogenic burst in the adult #DentateGyrus at P30 and ultimately results in a premature depletion of the NSC pool. Essentially, too much too soon leads to not enough later !!

2️⃣How did we figure this out? We used conditional mutant 🐁 during critical developmental stages & found that Sox5 is required to restrict the initial entry in #Quiescence of NSCs. Intriguingly, we discovered a critical window in the second postnatal week when NSCs build up a shallow quiescent state.
